Bitter Orange contains small amounts of alkaloids such as synephrine and octopamine, which are direct- and indirect-acting adrenergic agents.

Adrenergic agents with beta-agonist activity stimulate lipolysis and increase the resting metabolic rate. Proto-alkaloids found in bitter orange acts as α2 receptor antagonists.

α2 receptor activation results in activation of mechanisms associated with the “fight or flight” response, including decreased lipolysis and negative feedback on release of norepinephrine. However, when an antagonist binds the α2 receptor different effects are observed. α2 antagonists are shown to decrease negative feedback resulting in more secretion of norepinephrine, in addition they are also shown to block antilipolytic α2 receptors on adipocytes.

This effect acts twofold on lipolysis: first by blocking antilipolytic action, and second by increasing plasma levels of norepinephrine which act on β-3 receptors to activate lipolytic mechanisms.

These proto-alkaloids in combination with other adrenergic agents such as synephrine and octopamine has the potential to act synergistically to increase rates of lipolysis, sympathetic tone and may also increase cognitive awareness and arousal.
